It is well recognized that Halal food safety work needs contritbution of Halal scientific facilities for screening adulteration of "Haram" (unlawful ingredients according to Islam). This is due to complexity of modem food industry nowadays. Gelatin is known as the most frequently found Haram. Gelatin is a protein derived from hydrolyzed collagen found within mammalian tissues, especially from porcine and bovine. The first year of current project aims to sh1dy an existence of obscured gelatin in food products available in the market. Screening procedure generally employs colorimetric method according to AOAC for analysis of hydroxyproline. The results revealed that amongse 316 screened products, 28.1 % of 135 Halal-certified products contained gelatin in comparison to 23.8% of 181 uncertified products. However, limitation of colorimetric screening is unability in identifying the animal origin of gelatin or collagen. Therefore, LC/MS/MS method was developed for overcoming such weakness. A hydrolytic method was thus developed prior to detection with LC/MS/MS which finally found the marker ion of gelatin at 915 m/z and 1044 m/z. Under the ms / ms, the ion at 915 m/z fragmented into 342, 568, 681 and 897 m/z while the ion at 1044 m / z fragmented into 471, 697, 810, 897 and 1026 m/z. The marker ion found for bovine gelatin was 915 and 1044 111 / z but the intensity or S / N ratio were lower. However, the marker ion for fish gelatin was not found. This technique has been employed in screening 10 samples of food products containing gelatin, 8 samples were found to be similar to porcine or bovine gelatins and 2 samples were similar to fish gelatin. The gelatin in food products can be extracted using absoluted ethanol and acetic acid in this study. The marker ion of the gelatin used in this study can distinguish fish gelatin from the mammalian ones namely porcine and bovine with provision of satisfactory results.
