An analytical method was developed for determination of plaunotol in human plasma using solid phase extraction (SPE) and gas chromatography (GC). The plasma proteins in a 0.5 ml plasma sample were precipitated with 1.0 ml acetonitrile. The resulting solution was subjected to a Sep-pak C18 cartridge equilibrated with 40% acetonitrile, and then washed with 3.0 ml of 10% ethanol in water. 2.0 ml of absolute ethanol was used to elute plaunotol. The obtained solution was evaporated, and 0.5 ml of hexane containing 2.5 ppm n-octacosane was added. Plaunotol in the eluted solution was analyzed by GC using a cappillary column and 2.5 ppm n-octacosane as internal standard. The limit of detection and limit of quantition were found to be 2.0 ppm and 3.0 ppm, respectively. By spiking 5.0 to 20.0 ppm plaunotol into the plasma sample, SPE gave the plaunotol recoveries of 89.2 to 104.2% for the intra-day (five samples) and 87.4 to 100.3% for the inter-day (five days), which are in the acceptable range of 80.0 to 110.0% for AOAC international standard. In comparison with the maximum acceptable range of 6.3 to 8.2% relative standard deviation for 5.0-20.0 ppm spiked plaunotol, the precision values of SPE were obtained to be acceptable for the intra-day (1.1 to 5.0% RSD), while the inter-day precision values of 0.5 to 8.5% RSD were found to be slightly higher than the acceptable range. Therefore, GC with SPE could be used for determination of plaunotol in plasma.
