Hairy roots were induced aseptically by inoculation on stems and leaf discs of Datura metel Linn. with Agrobacterium rhizogenes strains A4, 8196, R1000 and 15834. Good results were obtained from induction of leaf discs by R1000, A4 and 8196 in the dark. When isolated and cultured hairy roots on different media in the absence of phytohormones, they grew rapidly with extensive lateral branches comparing to ordinary roots. Growth index of fresh weight of hairy roots grown in modified B5 medium was twice of the ones in modified MS. Some transformed plants also regenerated from hairy roots these plants were characterized by short internodes, wrinkled leaves and grew slowly. The presence of atropine in hairy roots were examined by TLC and the quantitative determination by HPLC. Atropine accumulation of three hairy root lines induced by A. rhizogenes strain A4 varied considerably having highest yield of 0.49 % DW. Whereas, the highest of three lines induced by R1000 was 0.56% DW. The accumulation of atropine in hairy roots was double that of ordinary roots and the yield of the dark grown hairy roots was generally higher than the light grown one.
